Beckman Coulter FC500 Analyzer
The Cytomics FC500 is a user-friendly analytical instrument with two laser excitation wavelengths, 488nm (air-cooloed Argon ion laser, 20mW output) and 633nm (air-cooled Helium-Neon ion laser, 20mW output), that can measure up to 5 colors simultaneously depending on which laser is turned on.
A continuous pressure is applied to the sample tube based on user selected flow rates. Three flow rates are available: low (around 15 µl/min), medium (around 30 µl/min), high (around 60 µl/min).
Particle size range to analyze is 0.5 µm to 40 µm diameter.
This cytometer has the additional capability of 'walk-away' operation. It has a carousel that may be loaded with up to 32 tubes each to be run automatically.
Two FC500 analyzers are available currently in EOHSI building. Facility personnel provides help with the instrument acquisition, data analysis, and consultation for experiment design. Training is also available to enable investigators, their staffs, and graduate students to run the analyzer by themselves. Please contact us.

Tips for preparation of samples:
- Cell concentration in each tube: about 1 million cells/ml
- Sample volume in each tube: 0.5 - 1 ml (minimum volume is 0.5 ml)
- Sample tubes must be polypropylene (12x75 mm round bottom polypropylene tube- BD Falcon Cat# 352002 for non sterile, or Cat# 352063 for sterile).
- All samples should be filtered with 40 µm nylon cell strainers (Falcon # 352340) prior to running to avoid clogging the machines.
- If you have samples stained with more than one fluorochrome per sample, we recommend you bring single color stained positive control samples (each fluorochrome individually) and unstained or negative control (isotype) cells for proper compensation.
- We recommend that samples are kept on ice or chilled until ready to run on the instrument.
